Genetic variation in brain waste-clearance gene shapes sleep-related Alzheimer risk
Researchers at Edith Cowan University found that different versions of the AQP4 gene alter how short or disrupted sleep influences early Alzheimer-related brain changes.
A team from the Centre for Precision Health at Edith Cowan University analyzed how the aquaporin-4 (AQP4) gene, which regulates fluid flow in the brain’s nightly cleaning system, interacts with sleep patterns. Thirteen common AQP4 variants were compared with participants’ reported sleep duration, time to fall asleep, brain imaging results and cognitive performance over time. Some variants were associated with accelerated grey-matter loss in individuals who reported shorter sleep, while other variants linked longer sleep latency to reduced brain volume.
The researchers caution that these findings are preliminary and call for larger, more diverse studies before recommending genetic testing. They propose clinical trials that incorporate genetic data to test whether improving sleep can mitigate inherited vulnerability to Alzheimer’s disease. The work was published in the journal Alzheimer's & Dementia.
