High Blood Sugar Fuels Sugar Coat That Shields Tumors From Immune Attack
Researchers discovered that elevated glucose levels can thicken a sugar-rich glycocalyx on cancer cells, helping them evade immune detection.
Scientists at Sanford Burnham Prebys and partner institutions found that high glucose conditions, mimicking hyperglycemia, cause cancer cells to build a thicker glycocalyx—a sugar-laden surface shield—especially when cultured in a medium that reflects human body nutrients and in stiff, tumor-like settings. The protein heat shock factor 1 (HSF1) was identified as a key driver of this process; cells lacking HSF1 did not develop the enhanced coating.
Experiments indicated that the fortified glycocalyx hampers immune cells from recognizing and killing the tumor cells. Targeting HSF1 could therefore diminish the shield and improve immunotherapy outcomes. The work links metabolic disorders such as type 2 diabetes to a mechanistic pathway of tumor immune evasion, offering a potential drug-discovery target.
