New injectable gel could keep osteoarthritis meds in joints for weeks

Researchers at the University at Buffalo have created a hydrogel that turns solid in the joint, slowly releasing drugs and improving lubrication for several weeks after a single injection.

Scientists at the University at Buffalo engineered an injectable hydrogel that transitions from a fluid to a semisolid, lubricating depot once inside a joint. The matrix incorporates biocompatible polymers and drug-loaded nanocarriers, enabling high concentrations of hydrophobic compounds to be retained and released over multiple weeks via diffusion and matrix relaxation. Validation using a SIRT6 activator demonstrated sustained local drug levels, and the system’s dual function as a viscosupplement could improve joint movement while delivering disease-modifying therapy.

By concentrating medication within the joint, the technology may lower the need for repeated invasive injections and reduce systemic side effects. Although initially targeted at knee osteoarthritis, the platform could be applied to post-traumatic OA, disc degeneration, rotator cuff issues, and other localized drug delivery challenges.
