Research watchdog uncovers dozens of aging studies using bacterial antibody by mistake

Independent biologist Sholto David reported that more than 50 cell-ageing papers used an antibody that targets bacterial β-galactosidase instead of the intended mammalian protein, raising concerns about data reliability.

Sholto David, an independent molecular biologist, posted on July 21 that over 50 recent studies of cellular ageing employed an antibody designed for Escherichia coli β-galactosidase rather than the mammalian version used to mark senescent cells. He traced the mistake to papers that listed the bacterial reagent, which cannot reliably detect the enzyme in mouse or human tissues. The mis-selection highlights a broader challenge in biology, where insufficient validation of antibodies leads to irreproducible results, according to biochemist Aled Edwards and cancer researcher Jennifer Byrne.

One acknowledged case involved a 2010 Cancer Research article where Dan Liebermann mistakenly used the bacterial antibody, though he claims the error does not alter the paper’s conclusions. Journals including Cell, Nature Aging and others have been notified, and Tim Kersjes of Springer Nature said the publisher will assess the concerns and take editorial action if needed. The episode underscores the risk that flawed reagents could mislead future AI-driven drug-target predictions.
