Researchers identify natural mechanism that halts inflammation and eases pain
Scientists at University College London have discovered a fat-derived molecule pathway that speeds the resolution of inflammation and reduces pain in human volunteers.
University College London researchers have mapped a natural anti-inflammatory pathway involving small fat-derived molecules called epoxy-oxylipins. By blocking the enzyme soluble epoxide hydrolase with the drug GSK2256294, they increased these molecules in volunteers who were given a harmless injection of UV-killed E. coli to trigger a short-term inflammatory response. Both preventive and therapeutic dosing reduced intermediate monocyte numbers and hastened pain resolution, though external inflammation signs remained similar.
Laboratory work showed that one epoxy-oxylipin, 12,13-EpOME, suppresses the p38 MAPK signaling cascade that drives monocyte activation. The findings suggest a new route for safer treatments of chronic inflammatory diseases such as rheumatoid arthritis, without broadly suppressing immunity.
