Researchers uncover lymph-node-like immune hubs in mouse skull bone marrow

Scientists at Washington University School of Medicine identified previously unknown immune structures in mouse skull bone marrow that react to brain tumors before distant lymph nodes.

A study by researchers at Washington University School of Medicine in St. Louis revealed organized immune “security stations” in the bone marrow of mouse skulls that behave like lymph nodes. These hubs detected glioblastoma tumors before peripheral neck nodes and, when impaired, allowed faster tumor progression and earlier death in the animals. Conversely, boosting the skull marrow with immune-activating proteins helped mice reject tumors and live longer.

Although comparable immune cells were found in human skull marrow, the investigators have not yet shown that they form analogous structures or operate similarly in people. The discovery adds a new layer to the evolving view of brain-immune interactions, suggesting the skull may host dedicated immune surveillance sites. Future work must confirm the presence and role of these hubs in humans and explore how they might be targeted without provoking harmful inflammation.

Why it matters

If similar skull immune hubs exist in humans, they could become a novel target for treating brain cancers and other neurological diseases.
