Skull Bone Marrow Hosts Immune Hubs That Boost Brain Cancer Defense in Mice

Researchers identified hidden immune structures in mouse skull bone marrow that act as rapid responders against brain tumors, improving survival when enhanced.

Scientists at Washington University School of Medicine have uncovered lymph-node-resembling immune structures hidden in the skull bone marrow of mice, which serve as first-line defenders against brain tumors. Using a glioblastoma mouse model, they showed that interfering with these hubs hastened tumor progression, whereas strengthening them with a gel delivering three immune-boosting proteins triggered a rapid, localized antibody response that curtailed tumor growth and extended survival.

The immune activity originated in the skull hubs before spreading to external lymph nodes. Parallel analyses revealed comparable immune cells in human skull marrow, suggesting the mechanism may operate in people. The findings challenge the notion of the brain’s immune isolation and open avenues for therapies that target the skull directly for cancers and other neurological diseases. Senior author Jonathan Kipnis emphasized the potential to treat conditions like Alzheimer’s or Parkinson’s by accessing these “security stations” without systemic side effects.
