Sleep micro-awakenings may signal heightened Alzheimer’s genetic risk in middle age
Researchers found that frequent brief awakenings during sleep are linked to higher Alzheimer’s genetic risk in healthy middle-aged adults.
Scientists at the University of Liège, supported by the Stop Alzheimer’s Foundation, examined sleep recordings from more than 500 healthy individuals and calculated each person’s polygenic Alzheimer’s risk. In the subgroup aged 50 to 69, a higher frequency of brief nighttime micro-awakenings correlated with elevated genetic risk, while no such link appeared in participants aged 18 to 31. The researchers also investigated the locus coeruleus, a minute brainstem area that regulates wakefulness and REM sleep, noting its early involvement in Alzheimer’s pathology.
Findings, published in the journal Sleep, propose that subtle sleep disturbances might serve as early indicators of vulnerability before any memory loss emerges. The authors caution that the genetic scores remain low and cannot predict disease in any single person, but they see potential for sleep metrics to complement other screening methods. Future work will explore whether improving sleep quality could mitigate risk in genetically predisposed individuals.
